Is it possible to calculate area in AutoCAD drawings?

Firstly you should have the object in pline. @ coomand type Area> Select the object > Area will be dispalayd base on the units selected for drawing

Yes, with help of AREA command, Inside this command, there are sub-commands to Add or Subtract the Area as well, So that one can calculate Total Area.

There is enquiry command area. you can use it to check area by adding and subtracting. or Temporarily hatch the area for which you want to find area then select that hatch and go to properties palette and there palette shows the area that is covered by hatch. Hope that helped. Bhavin Patel

before that let me clarify u that area can be calculated only to the closed boundries, as well to open boundry is marked as perimeter and coming to commond (aa) AREA command, there are sub options to Add or Subtract the Area as well, So that one can calculate Total Area or to deduct the area.
