Stains used for light microscopy are colored molecules that bind to cell componets, affecting the light oassing through. The stains used for electron microscopy involve heavy metals taht affect the beams of electrons passign through.

Light microscopy involves coloured molecules and uses light as source. Whereas in Electron microscopy electric charges (electrons in particular) are obtained upon bombardment.

For instance in a red stain , it absorbs all other colors except red. The red light reaches to our eye. In electron microscopy, the source of illumination is electrons. The pattern of the electrons that passes is making the image . So sample must conduct electrons. So a stain comprising of heavy metal such like uranium is used for this puropose. read less
