Light microscopy involves coloured molecules and uses light as source. Whereas in Electron microscopy electric charges (electrons in particular) are obtained upon bombardment.

The pattern of the electrons that passes is making the image . So sample must conduct electrons. So a stain comprising of heavy metal such like uranium is used for this puropose. read less
