Yes. Grammar is very important for speaking too. Without grammar you can't make use of sentences. It's all like spoken English, You can't speak it without using grammar. Similarly you can't speak Sanskrit without grammar. It is the main root to the language.

Speaking Sanskrit will make you apply the grammar. Grammar is theory while speaking is the application or product of that theory. You can start speaking Sanskrit without knowing Grammar, as any other languages are learnt. But basic knowledge of grammar is indeed important.
