No, it is not necessary, but of course, if you know any instrument, it helps you as a reference. Sometimes you may not understand the exact frequency of a particular note; in that case, you can use any of your instruments to listen to that specific note again and again as a reference. But it is possible to learn vocal music without an instrument.
