Should I keep each Python class in a separate file?

Putting every separate class in a separate file discourages reading. This goes against the core philosophy of Python. Look at the structure of the Standard Library, the vast majority of modules are single-file modules, not packages.

Main problem statement : import a .xlsx file in python with a dataset value then after import we need to ensure that every time I enter a value in a row ; row must be added by 1 and automatically value changes to a row using python script without manually intervention . 
Use read_csv in a loop, there's no other way for this.

which is best in Rpa and python
Python is much preferrable, I would suggest you to go for Python. As it is now a days used in Automations too.
