Brief Description 1. Comparative inactivation of faecal indicator bacteria in response to environmental stresses The research work carried out during doctorate degree investigated the effects of various factors, namely exposure to sunlight (solar photo-oxidation), high temperature, chlorine, low pH, or starvation, on the enumeration of faecal indicator bacteria in water. The inactivated faecal indicator bacteria were enumerated on non-selective* & different selective** media under conventional aerobic conditions & under conditions designed to neutralise reactive oxygen species (ROS) to investigate the effects of the nutrient status and selectivity of the medium on bacterial counts. 2. Comparative inactivation of faecal indicator bacteria in traditional brass and earthern water storage vessels On the basis of this approach, the effectiveness of storing water contaminated with faecal indicator bacteria in brass or earthern vessels (mutkas) of the type used in rural India was investigated. In all cases results indicated that, while storage of water in a brass mutka can inactivate E. coli and coliforms over a 48 h period, standard aerobic plate counting using selective media may not be fully effective in enumerating sub-lethally damaged bacteria (Work was cited in Nature, New Scientist, Science Magazine, the Journal, Times of India, the Hindu etc). 3. Resuscitation and pre-enrichment of faecal indicator bacteria enumerated from environmental water samples Them-T7 agar was used for the resuscitation of total coliforms present in environmental water samples and its performance was compared with various selective media under aerobic and ROS-neutralised conditions. This resuscitative medium recovered more coliforms in 60% to 80% of samples compared to MacConkey agar under both sets of growth conditions, while all of the other selective media i.e. mFC-R agar, m-Endo agar and mLSA agar gave consistently lower counts after screening 50 water samples for total coliforms. 4. Development of a novel broth-based medium for the enumeration of injured E. coli The information gained was used for the development of a novel broth-based medium for the enumeration of sub-lethally injured Escherichia coli under field conditions. 5. Evaluation of a novel broth-based medium for the enumeration of Escherichia coli under field conditions The novel broth-based field medium (‘Coliblack’) was tested against a currently used method (H2S test) & evaluated under field conditions with unskilled personnel in rural locations in India. *Non-selective Escherichia coli- nutrient broth and agar; Enterococcus faecalis- brain-heart infusion broth **Selective Escherichia coli- MacConkey agar Chromocult Coliform agar, m-Lauryl sulphate agar, m- Faecal Coliform agar without rosolic acid, m-Endo Agar; Enterococcus faecalis- MacConkey agar no.2, Bile aesculin agar, Slanetz and Bartley, KF Streptococcus. (b) Research Summary as Research faculty in Lovely Professional University Independent research studies were carried out to evaluate the following areas: 1. Survival and detection of faecal indicator bacterium Enterococcus faecalis in drinking water samples exposed to sunlight (solar disinfection). 2. Survival and detection of faecal indicator bacterium Escherichia coli in drinking water samples exposed to sunlight (solar disinfection). Aim: The research focused to improve our understanding of the process by which Enterococcus faecalis and Escherichia coli is inactivated in sunlight & to apply the information gained to improve the methods available for detecting & enumerating this pathogen in drinking water samples collected from different sources in Lovely Professional University, Phagwara, India. Summary The study investigated the physiological processes underlying the inactivation of Enterococcus faecalis and Escherichia coli, as model organisms for water-borne bacterial diseases, in sunlight. Regular monitoring of drinking water sources for faecal indicator bacteria is an essential part of the procedures involved in monitoring the quality of water supplies. However, these bacteria are commonly found in samples of contaminated water where they undergo a variable amount of growth, injury or inhibition, due to their exposure to different biological and physico-chemical factors. These injured bacteria may lose the ability to grow on growth media in routine use, for example selective media, which are otherwise satisfactory for cultivation of healthy cells. The difference obtained in bacterial counts between non-selective and selective media has been used to describe the extent to which such bacteria have been subjected to stress. Stress is a physiological response produced in a bacterial cell as a result of a stimulus or because of a stressor. This stimulus or stressor, for example irradiation by short wavelength UV-A rays of sunlight, antimicrobial chemicals such as chlorine, heat, or cold, metals such as brass, etc. are predominantly present in natural habitats such as the environmental waters into which faecal indicator bacteria are discharged in sewage. The detection of these stressed bacteria using conventional (selective) culture procedures might lead to under estimation of their actual levels in environmental samples such as drinking water, resulting in a significant problem in accurately estimating their numbers. One of the most important factors responsible for the inactivation of bacteria in natural waters is sunlight, & this knowledge has been utilised in a process termed solar disinfection, whereby small amounts of contaminated water are exposed to strong sunlight for several hours prior to drinking. Solar disinfection is used on a global scale for the household-level treatment of drinking water under conditions where no alternative means of treatment is available (work carried out by Dr Rung a PhD student of Prof. Robert H Reed in University at Northumbria, New Castle upon Tyne, UK). The present research was extension of work carried out at Northumbria University which has demonstrated that enumeration of sunlight-damaged E. coli is enhanced under conditions where reactive oxygen species are neutralised. Further improvement was achieved by careful choice of the selective agents used in enumeration media, & similar processes were involved during exposure of Enterococcus faecalis and Escherichia coli to sunlight, providing scope for useful experimentation & careful selection of culture media.
